A plasmid miniprep template that adapts to your lab's vectors.

This plasmid miniprep template ships with a 10-step silica column protocol, validated kit references, pre-wired yield + purity QC fields, and structured inputs for source culture, plasmid identity, elution volume, eluate concentration, and A260/A280 ratio. Your lab edits the configuration to match the strain and vector you actually use, instead of rewriting the protocol every time you pull a new plasmid prep.

Template detail

What this plasmid miniprep template ships with.

This plasmid miniprep template ships configured for small-scale isolation of pUC19-GFP plasmid DNA from a 5 mL overnight DH5α culture using a silica-membrane spin column workflow, with a 10-step protocol covering harvest through Qubit-confirmed elution. The protocol assumes a Qiagen QIAprep Spin Miniprep Kit (or equivalent), alkaline lysis with P1 (resuspension + RNase A) / P2 (lysis) / N3 (neutralization) buffers, PB binding, PE wash, and elution in EB buffer or nuclease-free water.

Structured fields capture culture, lysis, and elution metadata in typed inputs (host strain, plasmid identity, culture volume, lysis buffer set, elution buffer, elution volume, eluate concentration, A260/A280 ratio), so prep outcomes are queryable across minipreps instead of buried in free text. Kit lots, column batches, and plasmid stock references link to the ELabELN inventory so reagent traceability flows into the experiment record without manual entry. The spreadsheet editor captures per-colony concentration and purity readings with calculated total yield, supporting yield-trend analysis across colony picks and strain backgrounds.

The template is intended as a tested baseline for academic and life sciences research labs running plasmid prep for restriction digest verification, Sanger sequencing, transfection, or downstream cloning steps. Configure the host strain, swap the plasmid identity, change the kit chemistry, adjust the culture volume and elution parameters, or move from low-copy to high-copy preparation by editing the culture section. ELabELN's tamper-evident audit trail captures every prep run, every QC reading, and every electronic signature, so the resulting record supports reproducibility documentation, multi-PI collaboration, and FDA 21 CFR Part 11 review when the lab's quality system requires it.

Frequently Asked

Common questions

Can I configure the template for my strain and plasmid?

Yes. Every section is editable. Swap in different host strains (DH5α, TOP10, JM109, Stbl3, XL1-Blue), change the plasmid identity, adjust the culture volume from 1.5 mL to 10 mL, switch between kit vendors (QIAprep, Promega Wizard, Zymo, NEB Monarch), or move from miniprep to midiprep by editing the culture and elution sections. Save your edits as a private template scoped to your lab, or publish back to the ELabELN template library.

How does the template differ from the DNA Extraction template?

This template covers plasmid DNA isolation from bacterial cells (circular DNA, alkaline lysis chemistry, typically 5-20 µg yields). The DNA Extraction template covers genomic DNA from mammalian cells (linear DNA, Proteinase K + ethanol binding, typically 1-5 µg from 1×10⁶ cells). Both use silica column chemistry but the buffer systems, lysis methods, and expected yields differ.

Does the template track yield + purity across colony picks?

Yes. The spreadsheet editor captures concentration (NanoDrop or Qubit) and A260/A280 + A260/A230 purity ratios, with total yield calculated from elution volume. Because the typed fields are queryable across preps, your lab can pull every miniprep from a specific colony or plasmid prep and compare yield and purity, supporting decisions about which colony to scale up for midiprep.

Is it included in every ELabELN plan?

Yes. Every published template in the ELabELN library is available to all ELabELN subscribers, with unlimited users.

Can my whole lab use this template?

Yes. ELabELN includes unlimited users, so the PI, postdocs, grad students, rotating researchers, and visiting collaborators can all use the template without per-seat charges. Granular permissions let the PI control who can edit preps versus only review them.

How quickly can my lab start running minipreps in ELabELN?

ELabELN Standard cloud deployment is typically live in 1-2 days. Once the instance is provisioned, this template loads from the library in a single click, with the strain, plasmid, and prep parameters editable to match the preps your lab actually runs.
